Investigation of Osteopontin Gene Polymorphism in Individuals with Chronic Periodontitis

PURPOSE: Periodontal diseases are infectious diseases characterized by tissue structure and destruction. Various studies have described the effects of different genotypes of cytokines and tissue structures that affect the prognosis of periodontal diseases. The inter-individual variability in the rate of destruction and prognosis of periodontal diseases may be associated with genetic differences. In recent years, it has been reported that the protein encoded by the OPN gene plays a role in inflammation, fibrosis and bone destruction mechanisms. In this study, the genotype distribution, allele frequency and genotype-phenotype relationship of the OPN gene in periodontal diseases were investigated.

METHOD: The severity of periodontal destruction was determined by performing periodontal clinical (probing pocket depth, attachment value, probing bleeding and plaque index values) and radiographic examinations in 50 healthy and 50 chronic periodontitis patients participating in the study. Genomic DNA obtained from peripheral blood samples taken from patients was also studied for 8090 C/T polymorphism in the 6th exon of the OPN gene by the PRC-RFLP method. The data of the study were analyzed statistically using prametric and non-prametric methods.

CONCLUSION: OPN gene +8090 C/T polymorphism may be one of the genetic factors that may predispose to chronic periontitis. It is necessary to conduct studies on this subject that will include more patients.
